从噬菌体展示随机肽库中淘选多肽药物

以凝血酶为靶分子,利用噬菌体展示和亲和淘选技术,从随机十五肽库中筛选到结合凝血酶的3个特异结合肽克隆,并对其中结合活性最强的短肽进行了序列测定。ELISA鉴定结果表明,3个克隆对于凝血酶都有一定的结合能力,且都与凝血酶的天然抑制剂水蛭素产生竞争。

Abstract
Thrombin as target molecule, and with the new bio-technique of phage display and biopanning, 3 special binding peptide clones were selected out from a phage-displayed random 15-peptide library. The sequence of the clone, which had the highest affinity with thrombin, has been determined. Data of ELISA showed that all the three clones could be combined to thrombin to some extent. And all of them could compete with hirudin, which is the natural inhibitor of thrombin.
