有关病毒poly（A）尾巴问题

我看了一些资料说，一些病毒属（比如马铃薯Y病毒属）介绍有poly（A）尾巴，但我查了这些属的病毒全基因好像没有poly（A）尾巴，这是怎么回事啊，我用oligo（T）15做RT-PCT没有结果，而用特异引物有结果，是不是问题出在这里呢？
  请各位大侠多多指点……

应该是正链RNA病毒有polyA，负链RNA病毒没有！

应该有的。有些时候体外转录病毒的侵染性克隆的时候还特地需要加A。。。

There is poly A tail! But nobody will put the a tail to the databank, I think this is because nobody knows how many A are there in the virus genome.

I have used oligo d T to amplify several potyviridae virus, it works fine without any problem.

The reason why you haven't get the product maybe you have the bad cDNA although you could amplify with the specific primers. I have the experience that if I do the RT using RANDOM primer, it is difficult to get product using  oligo d T.
