    新华网伦敦１月５日电（记者葛秋芳）英国一家公司日前宣布研制出一种控制流行性感冒的新型疫苗，这种疫苗在首次人体试验中取得了较好效果。

    英国ＡＣＡＭＢＩＳ公司发布的消息称，这种名为ＡＣＡＭ－ＦＬＵ－Ａ的疫苗，理论上具备预防所有Ａ型流感病毒毒株的潜力。与现有流感疫苗不同，新疫苗不需要每年随
着流感病毒变异而改变配方，也不必非用受精鸡蛋（鸡蛋胚胎）制造，只要通过细胞培养就能生产。

    新疫苗主要针对Ｍ２Ｅ，这是存在于所有Ａ型流感病毒毒株表面的一种小蛋白质。试验结果显示，疫苗对人体是安全的，接种了疫苗的志愿者能够产生针对Ｍ２Ｅ的免疫反应。

    为提高疫苗效力，研究人员还将其与一种被称为ＱＳ－２１的佐剂联合使用，这种佐剂由免疫抗原制成，能增强免疫系统活力并提高其对病毒的识别能力。试验结果发现，志愿者在接种添加了佐剂的疫苗后，有９０％的人体内产生了针对Ｍ２Ｅ的抗体。

    研究人员还对较容易感染禽流感和人类流感的白鼬进行了研究。他们将白鼬分成两组，给其中一组接种新疫苗，而另一组不接种，然后让它们分别接触大剂量的Ｈ５Ｎ１型禽流感病毒。研究结果表明，没接种疫苗的白鼬全部死亡，接种了新疫苗的白鼬７０％安然无恙。

    ＡＣＡＭＢＩＳ公司负责研发的执行副总裁迈克尔·沃森说，这种疫苗有可能在３年到５年内投放市场。

ACAM-FLU-A™ is a recombinant vaccine that uses a hepatitis B core protein (HBc) to present M2e, the extracellular domain of the ion channel protein M2(5). M2 is one of the three proteins expressed on the surface of the ‘A' strain influenza virus and of infected cells alongside haemagglutinin (HA) and neuraminidase (NA). Unlike HA and NA, M2 is highly conserved, albeit under natural conditions not easily recognised by the immune system. M2e could elicit an immune response to all influenza ‘A' strains. As such, ACAM-FLU-A™ has the potential to be both a universal pandemic or pre-pandemic influenza vaccine and part of a universal seasonal vaccine. Historically, influenza pandemics have been caused by ‘A' strains of the virus while seasonal vaccines target both ‘A' and ‘B' strains of the virus.

流感通用疫苗

英国Acambis公司称，“ACAM-FLU-A”疫苗应该能预防所有种类的A型流行性感冒，而且在人体试验上已经取得很好的结果。

目前，生产冬季流行性感冒疫苗的公司必须定期重新设计疫苗，因为感冒病毒不断地演变。

新的疫苗就能克服这个问题，而且能事先大量制造储存起来，应付禽流感暴发。

在英国，每年冬天有大约4000人死于感冒以及感冒所引起的各种并发症。

以全球来算，每年有介于50万至100万的人死于流行性感冒。

但是科学家相信，如果全世界发生人传人的禽流感，最多可能会夺走5000万人的性命。

在美国进行的人体试验显示，这种流感疫苗对人体是安全的，能够快速地让人体对感冒产生免疫力。

注射两个剂量的ACAM-FLU-A的人，10个就有9个人产生抗体，能抵抗感冒病毒。

Acambis公司正在进一步试验，准备找出最佳化的疫苗组成成分，然后再进行较大规模的人体试验。

3，There is a high expression  of  M2  in  the surface of virus- infected cells
4，Possess ion channel activity ,and  a role for M2 in assembly and budding (some other functions）
5，Maintains a highly conserved sequence over long periods of time .（1918-2005）
6,M2e-specific Abs have been shown to display significant  protective  activity  in animal  models: homologous    and heterosubtypic  viruses challenge

M2 vaccine candidates that have been explored include peptide-carrier conjugates , baculovirus-expressed M2 , fusion proteins , multiple antigenic peptides , and M DNA constructs that potentially  express M2 .不过个人认为：针对表面保守蛋白M2胞外区（M2e)的疫苗应该和针对中和抗原HA疫苗联合使用会更好，毕竟HA抗体是流感病毒诱导产生的唯一的中和抗体，其在预防流感病毒感染中的作用是无法代替的哦！
